It is much worse that NotesDatabase class does not have an IsExist method.
Well, the appropriate functions exists in NotesDatabase:
NotesDatabase.IsOpen and Notesdatabase.Close()
But I guess you want to check, if somebody else is using the database (isOpen), or block it from usage (Close)? This can only be done using administrative console commands.
